In this experiment, the released sample was measured at different time points using HPLC. Fig. 2c shows the release of DOX from the self-assembled nanoparticles at 37 °C in different PBS media (pH 7.4 and pH 6.0), indicating desirable stability of the DOX@TPP–PEG–biotin SANs. After 24 h, the DOX released was only 30.0 ± 1.8% in the pH 7.4 PBS medium compared to that in the pH 6.0 PBS medium (65.4 ± 2.0%). These results indicate that under acidic conditions the DOX was released into the cellular medium. The sizes and zeta potentials in Fig. 2a and b showed no aggregation behavior at room temperature for five days at 37 °C, indicating that the self-assembled nanoparticles prepared in this study showed adequate stability.
